Whenever I visit a city for the first time, I try to do three things.

#1) Take a food tour focusing on local culinary traditions - especially if the trip is longer than a weekend. Not only do you learn a lot about the destination, the guide usually has fantastic restaurant recommendations.
#2) Wake up early and walk the city streets as the sun rises. There is nothing like experiencing the empty streets and watching the city awaken. One of my favorite memories of Rome was a Sunday morning, the scent of garlic and tomatoes filling the air as the nonnas started their Sunday cooking.
#3) Take a bike tour - You can cover more ground than on a walking tour and you get much more of the city atmosphere than you do on a tour bus.
